Abstract
Various supercritical particle formation processes have been employed to develop bioactive delivery systems with the resulting nano/microparticles having morphologies and functionalities superior to those generated by conventional techniques. Several processes have been developed in our lab, targeting the delivery of hydrophilic and hydrophobic bioactives. Solid lipid particles were formed and loaded with lutein or vitamin B2 upon rapid depressurization of the CO2-saturated lipids into air or water environment, respectively. Liposomes were prepared by pressurization of soy lecithin + bioactive dispersion in water with CO2 followed by rapid depressurization. Nano-scale unilamellar vesicles of liposomes loaded with bioactives (lutein, co-enzyme Q10, anthocyanin) with a narrow particle size distribution were obtained. Polysaccharides (oat beta-glucan, gum arabic, alginate) and proteins (soy, egg white and whey protein) were dried using the PGX (Pressurized Gas-eXpanded liquid) technology and polysaccharides were loaded with bioactives (CoQ10, vitamins D and E) using adsorptive precipitation. Controlling the structure of all these soft particles to achieve the targeted functionality relies on proper control of process parameters based on an understanding of the fundamentals.
